Regulators of cerebellar granule cell development act through specific signaling pathways.

چکیده
The proper development of the central nervous system depends upon a finely tuned balance between cell proliferation and programmed cell death (PCD). Although PCD was initially believed to depend solely on the inability of certain neurons to obtain access to a limited supply of trophic factors, it has become apparent that the local production of death signals is also critical. In this Viewpoint, we discuss several pathways implicated in the survival of cerebellar granule cells- both pathways that protect from apoptosis and pathways that promote apoptosis-and describe how these disparate pathways converge on the final common mediators of PCD. Information on other important pathways implicated in granule cell survival may be found in the Connections Maps.

The Time Course of JNK and P38 Activation in Cerebellar Granule Neurons following Glucose Deprivation and BDNF Treatment
Low glucose condition induces neuronal cell-death via intracellular mechanisms including mitogen-activated protein kinases (MAPK) signaling pathways. It has been shown that low glucose medium decreases neuronal survival in cerebellar granule neurons (CGNs). In this study, we have examined the activation of JNK, p38kinase and ERK1/2 pathways in low glucose medium in CGNs. The CGNs were prepared ...

متن کاملThe Jak-Stat Signaling Pathway of Interferons System: Snapshots
Interferons (IFNs) are a family of small regulatory glycoproteins that play a central role in the defense against viral infections. Although IFNs have been initially discovered as antiviral factors, today they are known as an integral part of the cytokine network that affect a wide range of biological processes. IFNs exert their pleiotropic effects through their multisubunit cell surface recept...
متن کاملSemaphorin-3F attracts the growth cone of cerebellar granule cells through cGMP signaling pathway.
Growth cone extension is guided by extracellular factors during the brain development but the underlying cellular mechanisms remain largely unclear. Here, we examined the potential function of class-3 semaphorins in cultured cerebellar granule cells. We found neuropilin-2 (NP2), the high-affinity receptor for semaphorin-3F (Sema3F), is highly expressed in cerebellar granule cells. An extracellu...
متن کاملHedgehog and PI-3 kinase signaling converge on Nmyc1 to promote cell cycle progression in cerebellar neuronal precursors.
Neuronal precursor cells in the developing cerebellum require activity of the sonic hedgehog (Shh) and phosphoinositide-3-kinase (PI3K) pathways for growth and survival. Synergy between the Shh and PI3K signaling pathways are implicated in the cerebellar tumor medulloblastoma. Here, we describe a mechanism through which these disparate signaling pathways cooperate to promote proliferation of ce...
